白色念珠菌檢查方法菌株培養(yǎng)：
1、增菌培養(yǎng) 取供試液10ml（相當于供試品1g、ml、10cm2）直接或處理后接種至適量（不少于100ml）的沙氏葡萄糖肉湯培養(yǎng)基中，培養(yǎng)48～72小時。
2、分離培養(yǎng) 取上述培養(yǎng)物劃線接種于沙氏葡萄糖瓊脂培養(yǎng)基平板上，培養(yǎng)24～48小時（必要時延長至72小時）。
結果判斷：
1、白色念珠菌在沙氏葡萄糖瓊脂培養(yǎng)基上生長的菌落呈乳白色偶見淡黃色，表面光滑有濃酵母氣味，培養(yǎng)時間稍久則菌落增大，顏色變深、質(zhì)地變硬或有皺摺。若平板上無菌落生長或生長的菌落與上述菌落形態(tài)特征不符，判供試品未檢出白色念珠菌。
2、如平板上生長的菌落與上述菌落形態(tài)特征相符或疑似，應挑選2～3個菌落分別接種至念珠菌顯色培養(yǎng)基平板上，培養(yǎng)24～48小時（必要時延長至72小時）。若平板上無綠色或翠綠色的菌落生長，判供試品未檢出白色念珠菌。
3、若平板上生長的菌落為綠色或翠綠色，挑取相符或疑似的菌落接種于1%吐溫80玉米瓊脂培養(yǎng)基上，培養(yǎng)24～48小時。取培養(yǎng)物進行染色，鏡檢及芽管試驗。
4、芽管試驗 挑取1%吐溫80玉米瓊脂培養(yǎng)基上的培養(yǎng)物，接種于加有一滴血清的載玻片上，蓋上蓋玻片，置濕潤的平皿內(nèi)，置35～37℃、1～3h，置顯微鏡下觀察，可見到由孢子長出短小芽管。
5、若上述疑似菌為非革蘭陽性菌，顯微鏡未見厚膜孢子、假菌絲、芽管，判供試品未檢出白色念珠菌。

Result judgment:
1, candida albicans in savary-gilliard glucose AGAR culture medium on the growth of colony has milk-white accidentally see pale yellow, smooth surface has a strong smell, yeast cultivation time slightly long increases colony, darker color, texture, harden or buckling. If there is no colony growth or growth on the plate and the above colony morphological characteristics are inconsistent, it is judged that no candida albicans has been detected in the test product.
2, such as tablet on the growth of colonies are in conformity with the colony morphology or suspected, should choose 2 ~ 3 colonies were inoculated to candida color medium plate, cultivate 24 to 48 hours (ShiYanChang necessary to 72 hours). If there is no green or greenish-green colony growth on the plate, no candida albicans was detected in the test product.
3. If the bacterial colonies on the plate are green or emerald green, select suitable or suspected colonies and inoculate them on 1% tween 80 corn AGAR medium for 24-48 hours. Culture materials were used for dyeing, microscopic examination and bud tube test.
4, 80 corn germ tube test pick 1% twain AGAR culture medium, inoculated on the slide with a drop of blood serum added, cover glass, in wet plate, buy 35 ~ 37 ℃, 1 ~ 3 h, microscope, visible to grow short and germ tube by spores.
5. If the above suspected bacteria are non-gram positive bacteria, no thick film spores, pseudomycelia, and germ tubes have been observed under the microscope, and no candida albicans have been detected in the test products.
